目的:检测DNA聚合酶ι(polymerase iota,Polι)基因在人肺癌组织、癌旁组织及正常肺组织中的表达,结合肺癌临床病理特征及预后分析Polι在肺癌发生、发展中的意义。方法:选取2001年1月至2003年8月在山东大学齐鲁医院胸外科行手术治疗的明确诊断为原发性肺癌的患者,应用免疫组化和RTPCR方法检测62例肺癌组织、30例癌旁组织和30例正常肺组织标本中Polι的表达水平,采用SPSS13.0软件对数据进行统计学分析。结果:人肺癌组织、癌旁组织和正常肺组织中Polι表达率分别为54.8%、333%和10.0%,不同类型肺癌组织中Polι表达无显著性差异(P>0.05),随着肺癌分化程度升高,Polι表达下降(P<0.05),Ⅲ期和Ⅳ期肺癌组织中Polι表达明显高于Ⅰ期和Ⅱ期(P<0.05),生存3年以下肺癌Polι表达明显高于3年以上者(P<0.05),吸烟者Polι表达高于非吸烟者(P<0.05)。结论:Polι高表达在肺癌发生、发展中起到重要作用,可能作为肺癌生物学特征和预后判断的参考指标。

[Abstract]
Objective:To investigate the expression of DNA polymerase iota (Polι) in human lung cancer tissues, adjacent lung cancer tissues, and normal lung tissues, so as to analyze the role of Polι gene in the development and progression of lung cancer. Methods: Samples were obtained from lung cancer patients who were surgically treated during Aug. 2003 to Jan. 2001 in Qilu Hospital of Shandong University. Immunohistochemisty and RTPCR were applied to detect Polι expression in 60 lung cancer specimens, 30 adjacent lung cancer specimens and 30 normal tissue specimens. Data were analyzed with PSS13.0 software. Results: The positive rates of Polι in lung cancer, adjacent lung cancer tissues and normal tissues were 54.8%, 33.3% and 10.0%, respectively. Polι expression levels were similar in different types of lung cancer (P>005). Polι expression in lung cancer tissues was decreased with the increase of differentiation grade of lung cancer (P<0.05), with the expression in grade Ⅲ and Ⅳ of lung cancer significantly higher than those in grade Ⅰ and Ⅱ(P<0.05). Polι expression in patients who survived less than 3 years was markedly higher than in those survived longer than 3 years (P<005); and smoking patients had a markedly higher expression than nonsmokers (P<0.05). Conclusion: High expression of Polι plays an important role in development and progression of lung cancer, and it may be used in evaluating the biological characteristics and prognosis of lung cancer.
