目的:探讨Toll样受体(Toll-like receptors,TLR)4和TLR9在胃癌组织中的表达及其与临床病理特征的关系。方法:收集2009年1月至2009年10月在解放军第174医院手术切除的42例胃癌及其癌旁(距癌灶边缘5 cm)组织标本。RT-PCR和 Western blotting检测胃癌组织中〖STBX〗TLR4〖STBZ〗、〖STBX〗TLR9〖STBZ〗在mRNA和蛋白水平的表达。结果:胃癌组织中〖STBX〗TLR4〖STBZ〗和〖STBX〗TLR9〖STBZ〗 mRNA表达高于癌旁组织(1.29±0.03 vs 0.53±0.02, 0.99±0.04 vs 0.22±0.05;均P<0.05),Ⅲ期+Ⅳ期胃癌组织中〖STBX〗TLR4〖STBZ〗和〖STBX〗TLR9〖STBZ〗 mRNA表达显著高于Ⅰ期+Ⅱ期(P<0.05),有远处转移的胃癌组织中〖STBX〗TLR4〖STBZ〗和〖STBX〗TLR9〖STBZ〗 mRNA表达显著高于无远处转移者(P<0.05); 〖STBX〗TLR4〖STBZ〗和〖STBX〗TLR9〖STBZ〗 mRNA的表达水平与患者的年龄、性别、淋巴结转移没有相关性。胃癌组织中TLR4和TLR9蛋白的表达高于癌旁组织(1.36±0.05 vs 0.48±0.04, 1.12±0.05 vs 0.34±0.03;均P<005),Ⅲ期+Ⅳ期胃癌组织TLR4和TLR9蛋白的表达显著高于Ⅰ期+Ⅱ期(P<0.05)。结论:TLR4和TLR9在胃癌组织中高表达,其表达与胃癌分期、远处转移具有相关性,表明TLR4和TLR9可能参与了胃癌的发生和发展,可能为胃癌的治疗提供一种潜在的靶点。

[Abstract]
Objective:To investigate the correlation of Toll-like receptors(TLR)4 and TLR9 expression with the pathological stages and metastases of gastric cancer. Methods: Forty-two gastric cancer and paracancerous tissues were included in the present study (patients were diagnosed in No.174 Hospital of PLA from Jan.1 2009 to Oct. 2009). The mRNA and protein expressions of 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 in the gastric cancer and paracancerous tissues were detected by RT-PCR and Western blotting analysis, respectively. Results: The expressions of 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 mRNA were significantly higher in the gastric cancer tissues than those in the paracancerous tissues (1.29±0.03 vs 0.53±0.02, 0.99±0.04 vs 0.22±0.05;P<0.05). T3+T4 gastric cancer tissues demonstrated significantly higher levels of 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 mRNA than the T1+T2 tissues (P<0.05). Significantly higher levels of 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 mRNA were observed in patients with distant metastases than in those without (P<0.05). However, no correlation was observed between 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 mRNA expression with the age, gender or lymphatic metastasis status of patients. The expressions of 〖STBX〗TLR4〖STBZ〗 and 〖STBX〗TLR9〖STBZ〗 protein were significantly higher in the gastric cancer tissues than those in the paracancerous tissues (1.36±0.05 vs 0.48±0.04, 112±0.05 vs 0.34±0.03;P<0.05), T3+T4 gastric cancer tissues demonstrated significantly higher levels of TLR4 and TLR9 proteins than the T1+T2 tissues (P<0.05). Conclusions: TLR4 and TLR9 are up-regulated in gastric carcinoma tissues, and TLR4/9 expression is correlated with the TNM staging and distant metastasis. These data suggest that TLR4/9 may be involved in the development and progression of gastric cancer, and TLR4/9 may be potential target for gastric cancer treatment.
