目的:探讨鸦胆子油乳在体外对人大细胞肺癌NCIH460细胞的抑制作用及其机制。方法:以不同质量浓度鸦胆子油乳(2.5、5.0、10、20、40、80 μg/ml)处理NCIH460细胞,24、48、72 h后收集细胞,MTT法检测鸦胆子油乳对NCIH460细胞增殖的抑制作用;吉姆萨染色后光学显微镜下观察NCIH460细胞形态学改变;流式细胞仪测定NCIH460细胞凋亡率;caspase3酶活性试剂盒检测NCIH460细胞caspase3酶活性。结果:鸦胆子油乳可剂量和时间依赖性抑制NCIH460细胞的增殖,80 μg/ml 鸦胆子油乳作用72 h时的抑制率达(91.07±1.60)%。鸦胆子油乳作用后,NCIH460细胞呈现典型凋亡形态学改变;流式细胞术检测结果显示,10、20和40 μg/ml鸦胆子油乳作用48 h后,NCIH460细胞凋亡率分别为(45.23±430)%、(54.14±3.09)%和(61.57±7.28)%(P<0.01)。鸦胆子油乳可剂量依赖性上调NCIH460细胞caspase3酶活性,40 μg/ml鸦胆子油乳时caspase3酶活性为(1.07±0.07)μmol/μg,是对照组的4.97倍(P<0.01)。结论:鸦胆子油乳可能通过活化caspase3诱导NCIH460细胞凋亡,抑制NCIH460细胞增殖。

[Abstract]
Objective: To investigate the inhibitory effect of Brucea javanica oil emulsion on the growth of human largecell lung cancer NCIH460 cells in vitro and its mechanisms. Methods: NCIH460 cells were treated with different concentration of Brucea javanica oil emulsion (2.5, 5.0, 10, 20, 40 and 80 μg/ml) for 24, 48, and 72 h. The inhibitory effects of Brucea javanica oil emulsion on proliferation of NCIH460 cells was examined by MTT; morphological changes was observed by optical microscope after Giemsa staining; apoptotic rate of NCIH460 cells was examined by flow cytometry; and caspase3 activity in NCIH460 cells was detected with caspase3 activity kit. Results: Brucea javanica oil emulsion significantly inhibited the proliferation of NCIH460 in a dose and timedependent manner, and the inhibiting rate reached (91.07±1.60)% after 80 μg/ml Brucea javanica oil emulsion treatment for 72 h. Typical apoptotic morphological changes also appeared after Brucea javanica oil emulsion treatment. Flow cytometry results demonstrated that the apoptotic rates of NCIH460 cells were (45.23±4.30)%, (54.14±3.09)% and (61.57±7.28)% after 10, 20 and 40 μg/ml Brucea javanica oil emulsion treatment for 48 h, respectively (P<0.01). In NCIH460 cells, the Brucea javanica oil emulsion increased the activity of caspase3 in a dosedependent manner. after 40 μg/ml Brucea javanica oil emulsion treatment, caspase 3 activity reached 1.07±0.07 μmol/μg, which was 4.97 fold higher than control group (P<0.01). Conclusion:Brucea javanica oil emulsion can induce apoptosis and inhibit the proliferation of NCIH460 cells, possibly through increasing caspase3 activity.
